Quick answer
Use the latest Chrome, Edge or Safari, make sure Snapchat on your phone is up to date and logged in to the same account. Snapchat says you can only be logged in on one computer at a time. For calls, allow microphone and camera permission, check nothing is muted (including the browser tab), switch sources with the up arrow next to the mic or camera icon, and disconnect headphones and Bluetooth.
